Abstract: Miconia ligustroides (DC.) Naudim belonging to the Melastomataceae family, is an endemic species of Brazil, which has analgesic, antimicrobial and is on the list of recommended species for restoration of riparian forests. The objective was to establish an in vitro germination and micropropagation protocols for M. ligustroides. The seeds were sterilized and inoculated into culture media, saline formulations based on MS and WPM medium supplemented with five concentrations of GA3 (0.0; 5.77; 11.54; 17.31; and 23.08 µM). Leaf explants and nodal root seedlings were inoculated on MS medium supplemented with different concentrations of BAP (0.0; 13.5; 27.0; 40.5; 54 and 135 µM) and 2,4-D (0.0; 4.5; 13.5; 22.6 and 45.2 µM) for inducing indirect organogenesis. For rooting of shoots MS medium supplemented with IBA (0.0 and 9.8 µM) was tested. Rooted shoots were acclimatized ex vitro in Tropstrato® substrate. Higher germination percentage was obtained with MS medium free of GA3 . The highest sprouting rate was observed when the leaf and nodal explants were cultured in 54 µM BAP and 13.5 µM 2.4-D, respectively, and in the presence of 135 µM BAP for root explants. Shoot rooting was obtained without supplemental IBA. The use of 9.8 µM IBA promoted a significant root lenght and no influence on root number. The micropropagation steps achieved in this study indicate that germination, shoot multiplication, in vitro rooting and acclimatization of M. ligustroides can be successfully obtained.
